Amplitude’s proprietary taRNA platform distinguishes itself by decoupling replicase and antigen-encoding components. This structural separation allows for finer control over formulation and dose optimization, potentially addressing manufacturing bottlenecks inherent in current RNA modalities. Under the terms of the deal, Amplitude retains responsibility for early-stage taRNA optimization and specific preclinical research, while Lilly gains exclusive rights to manage subsequent clinical development, regulatory filings, and commercial scale-up.
Amplitude Therapeutics Partners With Eli Lilly to Advance taRNA Vaccines
Eli Lilly and Boston-based biotech startup Amplitude Therapeutics have signed a strategic licensing agreement to develop trans-amplifying RNA vaccines. The partnership focuses on high-need infectious diseases, combining Amplitude’s modular RNA technology with Lilly’s global infrastructure to refine antigen design and lower effective dosage requirements.

Dr. Cory Sago, CEO of Amplitude, described the agreement as a validation milestone for the company’s in vivo RNA platform. The initial scope covers undisclosed targets, with options for Lilly to expand the pipeline by two additional programs. Founded in 2022 and currently part of the Lilly Gateway Labs community, Amplitude is backed by investors including ARCH Venture Partners and Newpath Partners, alongside non-dilutive funding from the Gates Foundation and CEPI.
